Synthesis of human corticotropinyl-thiolglycine and its specific conjugation to bovine serum albumin

Human corticotropin containing a thiolglycine residue at the carboxyl terminal (I) was synthesized by the solid-phase method. The citraconyl derivative of peptide I was coupled to either a model tetrapeptide or bovine serum albumin by reaction with silver nitrate/N-hydroxysuccinimide in water. The extent of reaction with bovine serum albumin was determined by radioimmunoassay, and the peptide-protein conjugate was shown to possess 12% of the steroidogenic activity of porcine adrenocorticotropin in isolated rat adrenal cells. Peptide I and its conjugate with the model tetrapeptide were fully active in the same system.     

Sensitive visualization of antigen-antibody reactions in dot and blot immune overlay assays with immunogold and immunogold/silver staining

The use of colloidal gold and colloidal gold followed by silver enhancement as marker for dot or blot immune overlays is described. Colloidal gold probes concentrating at the sites of immune reaction gradually develop a pinkish colour during incubation that can be seen with the naked eye. With a physical developer, very high sensitivity and contrast is obtained by silver precipitation on the gold marker. Comparison of the immunogold and immunogold/silver staining methods with the indirect, PAP and ABC immunoperoxidase methods demonstrates that colloidal gold probes are excellent markers for immune overlay assays.     

沸石对猪血管损伤止血的作用及对组织影响的初步研究

目的:研究沸石对猪大血管损伤出血的止血作用及其对局部组织的影响. 方法:健康家猪12只(12.89±1.16 kg)随机分为实验组和对照组,每组6只,利刀横断股动脉、静脉和部分股四头肌. 致伤后20 s实验组应用沸石粉止血,对照组采用纱布加压止血. 15 min开始输入乳酸钠平衡盐溶液共250 mL. 观察120 min内平均动脉压、出血量、死亡率、死亡时间. 切取血管断缘两侧组织,进行HE染色,观察组织的病理改变. 结果:实验组动物应用沸石后2 min止血成功,动物死亡率0%;对照组止血失败,动物死亡率100%. 实验组血管断缘0.5 cm内部分组织变性坏死,断缘0.5 cm以远组织未见明显病理改变. 结论:沸石能有效控制本模型中动、静脉的致命出血,显著降低死亡率;沸石放热导致局部组织热损伤,但不影响清创血管吻合.     

纳米氧化铁黄材料对小鼠血清生化指标的影响

目的观察纳米氧化铁黄材料对小鼠血清生化指标的影响。方法以5g/kg剂量的纳米氧化铁黄给小鼠一次经口灌胃,14d后处死,观察小鼠血清生化指标的变化情况。结果纳米氧化铁黄染毒小鼠除血清总胆红素(T-BIL)有比较明显的降低外(P<0.05),其余血清生化指标与对照组相比差异无显著性(P>0.05)。结论纳米氧化铁黄对小鼠血清生化指标无显著影响。     

Use of vacuum-assisted closure for healing of a persistent perineal sinus following panproctocolectomy: report of a case

Perineal sinus is a troublesome complication after proctectomy for inflammatory bowel disease or rectal cancer. The results from treatment with simple dressings with or without surgical debridement are suboptimal. Definitive management may require complex operations with muscular or musculocutaneous flaps. Vacuum-assisted dressings have been used successfully for management of chronic wounds and soft tissue defects. This technique was successfully used in the management of a perineal sinus, which occurred after proctectomy for rectal cancer developing on a background of inflammatory bowel disease and preoperative radiotherapy.     

青银注射液治疗上呼吸道感染风热证118例临床疗效观察

目的探讨青银注射液治疗上呼吸道感染风热证的疗效。方法以清开灵注射液为平行对照，采用分层区组、随机、盲法、多中心Ⅱ期临床试验。观察青银注射液静滴治疗上呼吸道感染风热证118例。结果显示两组体温恢复正常时间及综合疗效组间差别无统计学意义（P〉0.05）。两组终点记分与基线比较的差别均有高度统计学意义。特别是改善咽痛、四肢酸痛、鼻塞流涕症状治疗组明显优于对照组，经统计学处理有显著性差异（P〈0．05）。结论青银注射液治疗上呼吸道感染风热证是一种良好的选择。     

大鼠被动型Heymann肾炎模型建立与抗体剂量关系的探讨

目的:通过对被动型Heymann肾炎模型的建立与抗体剂量关系的研究,进一步探讨膜性肾病的发病机制。方法:成年雄性SD大鼠分为A、B、C 3组,分别经尾静脉注射抗FX1A抗血清1 ml/100g、0.5 ml/100g以及生理盐水1 ml/100g,1周后检测24 h尿蛋白定量、血清白蛋白、血清肌酐、血清三酰甘油以及总胆固醇,分别在普通光学显微镜和透射电子显微镜下观察大鼠肾组织的病理学改变。结果:被动型Heymann肾炎模型的建立与SD大鼠尾静脉注射抗FX1A抗血清的剂量存在正相关。结论:膜性肾病的发生可能与抗体剂量有关。     

纳米硒对小鼠移植瘤H22的生长抑制作用及其机制

目的：研究纳米硒对小鼠移植瘤H22的生长抑制作用及其作用机理，探讨纳米硒抗肿瘤治疗的可能性。方法：采用体内抗肿瘤法观察各组的抑瘤率；肿瘤组织切片HE染色、甲基绿-派洛宁染色、透射电镜观察其病理改变；免疫组化法检测Bax蛋白的表达水平；HE切片检测对胸腺脾脏的黟响。结果：纳米硒抑制小鼠移植瘤H22的生长、浸润，减少核分裂，增加凋亡，血管生成减少；电镜见肿瘤细胞的典型凋亡形态，免疫组化显示肿瘤组织的Pax蛋白表达上调，SI、TI无明显降低，脾小体、脾巨噬细胞数升高。结论：一定剂量的纳米硒可能通过增强免疫作用和上调Bax蛋白的表达诱导肿瘤细胞凋亡两种主要途径来实现抗肿瘤作用。     

烧伤创面换药操作痛对患者外周血C-反应蛋白的影响及意义

目的观察严重烧伤患者创面换药操作痛前、后外周血C-反成蛋白（CRP）的变化，探讨CRP可否作为烧伤创面换药操作疼痛的应激反应的监测指标。方法测定在创而换药中有明显操作疼痛的烧伤患者（n=30，A组）和在创面换药中无操作疼痛的烧伤患者（n=30，B组）在创面换药前、后CRP水平的变化，并与正常对照组（n=10，C组）进行比较。结果与C组比较，A、B2组换药前、后CRP均显著升高（P〈0．01）；A绍换药的CRP水平较B组明显升高（P〈0．01）；A组换药后CRP水平较换药前升高（P〈0．05），B组换药前、后CRP无明娃变化（P〉0．05）。结论烧伤创面换药过程中严重的操作疼痛可引起患者外周血CRP升高，CRP可作为烧伤创同换药操作疼痛以及烧伤患者伤后产生应激反应的监测指标之一。     

Umbilical hernia repair in children: is pressure dressing necessary

The use of pressure dressing to cover the sutured surgical wound is usually considered a routine conclusion to the repair of umbilical hernias in children. The wound is usually left dressed for a minimum of 5–7 days. The main purpose of pressure dressing is prevention of a hematoma formation. The aim of this study was to compare the surgical outcome after umbilical hernia repair in children when the wounds were covered using pressure dressing or left exposed without dressing after the completion of wound closure. Ninety-six patients with umbilical hernia repair were prospectively randomized to receive pressure dressing (n = 52) or have their wounds left exposed without any dressing (n = 44) after the completion of wound closure. None of the hernias were huge umbilical hernia and none required an umbilicoplasty. In the group who received pressure dressing, one patient developed wound infection 1.9% while no patients developed wound infection in the group who had their wounds exposed without any dressing. In children, there was no significant difference in terms of wound infection, hematoma or seroma formation and recurrence rate after applying pressure dressing or leaving the surgical wounds exposed without any dressing after completion of wound closure. Pressure dressing after umbilical hernia repair may be unnecessary.
